Genomic Response in Daphnia to Chemical Pollutants

چکیده
Chemicals released into the environment have a potential to affect on various species and evaluation of the impacts on ecosystems including aquatic organisms is urgent issue. In order to evaluate chemical effects on various species, it is important to understand their mode of action. Recent advances in toxicogenomics, the integration of genomics into toxicology is promising for understanding mode of action in responding to chemical pollutants. In this study, we applied the toxicogenomics to an aquatic organism, Daphnia magna. Among aquatic organisms, Daphnia magna has been used extensively in acute toxicity or reproductive toxicity tests. Although these types of tests can provide information on hazardous concentrations of chemicals, they provide no information about their mode of action. Thus we developed an oligonucleotide-based DNA microarray based on a Daphnia expressed sequence tag (EST) database and applied for ecotoxicogenomic assessment of Daphnia magna. The DNA microarray was used to examine gene expression profiles of neonate daphnids exposed to several different chemicals. Exposure to these chemicals resulted in characteristic gene expression patterns that were chemicalspecific. This result indicates that our newly developed DNA microarray can be useful for a mechanistic understanding of chemical toxicity on a common freshwater organism, Daphnia magna, and that the Daphnia genomics is useful for aquatic ecotoxicology.

A Bayesian model for the identification of differentially expressed genes in Daphnia magna exposed to munition pollutants.
In this article we propose a Bayesian hierarchical model for the identification of differentially expressed genes in Daphnia magna organisms exposed to chemical compounds, specifically munition pollutants in water. The model we propose constitutes one of the very first attempts at a rigorous modeling of the biological effects of water purification. We have data acquired from a purification syst...

Daphnia are already an established model species in toxicology. This freshwater crustacean is used commonly for environmental monitoring of pollutants around the globe and plays an important role in establishing regulatory criteria by government agencies (e.g., US EPA, Environment Canada organization for Economic Cooperation and Development, Environment Agency of Japan). Consequently, daphniids...
متن کاملEvaluation of silver nanoparticles toxicity in Daphnia magna: Comparison of chemical and green biosynthetic productions
Recently nanoparticles, particularly silver nanoparticles, are broadly used in industry, hence the contamination of the environment with AgNPs has caused considerable concern. In this study, the toxicity of biosynthetic nanosilver produced by two macroalgae: Sargassum boveanum and Ulva flexuosa extracts were compared with chemical nanosilver in Daphnia magna. Size and quality of nanoparticles e...
متن کاملChemical induction of colony formation in a green alga (Scenedesmus acutus) by grazers (Daphnia)
The green alga, Scenedesmus acutus, grows in culture in unicellular form, but it forms colonies (coenobia) when exposed for 48 h to a chemical released by the grazer Daphnia magna. The colony-forming response can be evoked only in growing cells. The Daphnia factor affects colony size but not algal growth rate. The minimum concentration of Daphnia factor that induces colony formation is equivale...

Recent characterization of the complete genomic sequences of multiple eukaryotes and prokaryotes is rapidly increasing our ability to understand how genome structure evolves and how gene expression and individual performance is modified in response to environmental challenges. These advances hold the promise of making a direct connection between the genome and the phenotype of organisms in popu...
